Duke Sucellus is the second most profitable desert treasure 2 boss, currently bringing in over 6 mil an hour if you get everything on rate, and if you have good stats and gear. It has lucrative drops, like the magus vestige, eye of the duke and the virtus robe top.
Guide Outline
If you would prefer to watch a video-version of this guide, I have put that below. Otherwise, continue scrolling to read the written-version of this guide.
Requirements

Jagex recently changed duke where now you can prepare for more fights by having a higher herblore level… With level 82 herblore, you can prepare for 4 fights at once.
To be able to fight him, you either need to be doing desert treasure 2 or have fully completed it. This is a quest with some of the highest requirements in the game, including 75 firemaking, 70 thieving and 60 runecrafting.
Its recommended to use melee, and having at least 80 attack and strength is essential.. Also, 70 defence and 70 prayer is very beneficial as well.
Having at least level 80 mining can also speed up kills at duke, and a higher agility level removes the need for stamina potions.
Getting To Duke

Duke is located in the ghorrock dungeon at weiss, and the fastest way there is to use the ring of shadows teleport to the dungeon, but to be able to use that teleport, you need a tablet drop from duke.
So the second fastest way there is using icy basalt to weiss and running to the boss arena.
An alternative way to get there is using the fairy ring code DKS and then taking the boat from relekka to weiss.
Dukes Mechanics

Now lets go through every one of dukes mechanics and how to avoid them…

To be able to fight duke in the first place, you need to prepare potions within the arena. Then you use 2 potions on duke and can fight him once.
But as i mentioned, with a higher herblore level you can prepare for more fights at once, where 62, 72, and 82 let you prepare for 2, 3 or 4 fights at once.

Preparing these potions involves firstly running to each edge of the arena, and bypassing the lazer shooters. You can simply skip over the tiles that the lazer comes out of by using run and going on these marked tiles. On both paths, theres also black shadows which spawn lightning and stun you, so avoid stepping on the shadowed tiles…

On each side of the arena, there are “arder” mushrooms on the left side, and “musca” mushrooms on the right side. Once you reach one side of mushrooms, you simply just click on them to retrieve them, and the amount of mushrooms required depends on your herblore level.
People with under 80 herblore require 3 of each mushroom per kill, whereas if you have 80 or above in herblore, you only require 2 mushrooms per kill.
On the way back, you also need to avoid all of those attacks and lazers that i mentioned earlier.
So you start on one side, then collect the mushrooms and run to the opposite side to collect those mushrooms…. You can 3 tick or 1.5 tick to collect the mushrooms faster to speed up kills, and this is done by starting a 3 tick action from 1 tile away and clicking to collect the mushrooms, then walking back and repeating.

Then, you need salt to finish off the potions, and to get salt, its located in the central area of the arena. Here you just mine with your pickaxe until you have enough salt, and this time it depends on your mining level. If youre below level 80 mining, you need to do 3 mining actions per kill, whereas above 80 mining, you only need 2 actions. You can again, 3 tick here, which speeds up the process slightly.
There are also vents around the arena which periodically change in a clockwise rotation, so dodge those as well while youre here.

Then to finish off prep, you head over to the big barrels near the entrance, and you click on them to start preparing the potions. And you can prepare both barrels there, and collect the potions once theyre completed.

Once youre prepared, you click duke 2 times to start the battle, and during the kill, duke will auto attack you, every 5 gameticks, and you have to step back on the same tick that duke attacks you otherwise you take full damage. While you can survive without it, stepping back will significantly reduce the damage of this attack. If youre in long-ranged distance from duke, he uses magic attacks and those hit hard even through prayer.

After 5 auto attacks, duke does his eye attack, where you need to hide behind one of the pillars to avoid damage.
Then after the eye attack, he does 3 regular auto attacks, then he does a vent attack where he spawns poisonous clouds on the vent nearest to you.

And the number of vent attacks depends on dukes health… if hes above 50% its only 1 vent that gets targeted, so you just move out of the way once.
But if duke is below 50 percent health, 2 vents get attacked with poison. So for this, you just move one by one over the vents.
Eventually, duke will do the pillar attack again, and once below 25 percent health, duke enters an enraged phase, where every attack is 1 tick faster, so now 4 ticks per attack.
Full Fight Runthrough
Now I'll take you on a full fight runthrough at duke so you can see what to do.
Gear & Inventory
Now for gear and inventory, and duke is weak to slash attacks, so having your best strength bonus boosting armour, along with a slash weapon is best.
This is an example minimum gear and inventory setup for duke, and being a monster weak to demonbane, you can bring an arclight or an emberlight to duke for decent damage.

At minimum you want a few strength boosting items, like a fire cape, fighter torso and barrows gloves, and youll notice that in your inventory, you should bring a picaxe and a pestle and mortar. And in your rune pouch you can bring runes for thralls if you have them.
This is an example mid tier setup at duke, with a bit more budget..

This includes the bandos tassets, faceguard and ferocious gloves…
And lastly, this is the max setup for duke, including the scythe, oathplate armour and top tier strength boosting equipment.
